Thermo Scientific Phusion High-Fidelity DNA Polymerase sets the "gold standard" for efficient PCR. Phusion DNA Polymerase has a mismatch rate that is 50 times lower than Taq and 6 times lower than Pfu, making it the best choice for cloning and other applications requiring high fidelity. Compared with other DNA polymerases, Phusion DNA Polymerase has stable performance, less time-consuming protocols (even in the presence of PCR inhibitors), and can achieve higher yields with lower enzyme amounts.

When using Phusion Hot Start II High-Fidelity DNA Polymerase, because Phusion DNA Polymerase reversibly binds to a specific Affibody ligand, which can inhibit DNA polymerase activity at room temperature, Therefore, no non-specific products are produced during amplification.
Affibody ligands can also inhibit the 3´→5´ exonuclease activity of the polymerase, preventing primer and template DNA degradation during reaction construction. At temperatures that promote polymerase activity, the ligand is released, allowing full activation of the polymerase. Phusion Hot Start II DNA Polymerase reactivates immediately at higher temperatures, so a separate activation step is not required in PCR protocols.

Key Features

• Reaction setup at room temperature
• No non-specific amplification and primer degradation during reaction setup
• Due to unique Hot start technology enables zero-time reactivation
• High fidelity (52X Taq)
• Shortens the extension time, thereby speeding up PCR (15-30 s/kb)
• Stable reaction, Little optimization required
• Generate more product with very low enzyme amounts
• Provide Green buffer for direct loading onto gel (F-537S or F-537L)

< b>Applications

• High-fidelity PCR
• High-throughput
• Difficult-to-amplify (high GC content) templates
• Obtain sequencing templates
• Multiplex PCR
• Long PCR
• Cloning
• Mutagenesis
• Microarray

Using Phusion DNA Polymerase
Phusion DNA Polymerase The annealing temperature design rules are different from those of many common DNA polymerases (such as Taq DNA polymerase). For best results, use www.thermofisher.cn/tmcalculator.
