Phusion Plus DNA polymerase is a Hot-start high-fidelity DNA polymerase using protein fusion technology and universal primer annealing capabilities. It is a Pyrococcus -like proofreading DNA polymerase fused to a processivity-enhancing domain, allowing Phusion Plus PCR Master Mix to generate PCR sequences with high accuracy, sensitivity, and inhibitor tolerance. Its novel reaction buffer contains a proprietary additive that stabilizes the primer/template duplex during the 60°C annealing step. This unique buffer formulation improves PCR specificity by eliminating the need to optimize annealing temperatures for each primer pair individually, helping to save time and avoid human error in PCR runs. In addition, with Phusion Plus PCR Master Mix and its universal annealing function, different PCR reactions can be cycled simultaneously using the same protocol.
Phusion Plus PCR Master Mix is inactivated at room temperature due to a hot-start mechanism mediated by the parent molecule, allowing for preparation and storage of PCR reactions prepared at room temperature. Enzyme activity is not fully activated until the predenaturation step.
