Thermo Scientific FastDigest Eco57I restriction enzyme recognizes the CTGAAG(16/14)^ site and uses universal FastDigest buffer at 37°C The cutting effect is better within 5-15 minutes. Iso-cleavage enzymes: AcuI, Bsp6II, BspD6II, BspKT5I, Eco112I, Eco125I, FsfI, SstE37III.

Thermo Scientific FastDigest Eco57I is a high-end series of fast restriction enzymes that are 100% active in both universal FastDigest and FastDigest Green reaction buffers.

Universal buffer allows rapid single, double or multiple enzyme digestion of DNA in 5–15 minutes without the need for buffer changes or subsequent DNA purification steps. See FastDigest Enzyme Reaction Conditions table for enzymatic activity, heat inactivation, and incubation of this and other FastDigest restriction enzymes time. DNA modifying enzymes (such as Klenow fragment, T4 DNA Ligase, alkaline phosphatase and T4 DNA Polymerase) are 100% active in FastDigest buffer. Therefore, enzymes used in downstream applications can be added directly to the FastDigest reaction mixture.

For convenience, FastDigest Green buffer includes 1 density reagent and 2 tracking dyes for loading digestion reaction products directly onto the gel.

Note: FastDigest Eco57I requires only Mg²⁺ for its activity but is stimulated by SAM. 0.01 mM SAM increases FastDigest Eco57I activity more than 100-fold. Despite this, FastDigest Eco57I still struggles to completely cleave some substrates. Cleavage with FastDigest Eco57I requires at least two copies of its recognition sequence.

FastDigest Green Buffer provides the same high performance as colorless FastDigest Buffer in DNA digestion reactions and downstream applications. For applications requiring product analysis by fluorescence excitation (eg determination of concentration under UV light), the use of colorless FastDigest buffer is recommended.
