Thermo Scientific FastDigest CseI is one of a series of high-end fast restriction enzymes that are 100% active in both Universal FastDigest Buffer and FastDigest Green Reaction Buffer.

Universal buffer can quickly complete DNA single, double or multiple enzyme digestion in 5–15 minutes, without the need to change buffers or subsequent DNA purification steps. For a table of enzymatic activities, heat inactivation, and incubation times for this and other FastDigest restriction enzymes, see Reaction conditions for FastDigest enzyme< /a>. DNA modifying enzymes (such as Klenow fragment, T4 DNA Ligase, alkaline phosphatase and T4 DNA Polymerase) are 100% active in FastDigest buffer. Therefore, enzymes used in downstream applications can be added directly to the FastDigest reaction mixture.

For convenience, FastDigest Green Buffer includes 1 density reagent and 2 tracking dyes for loading digestion reaction products directly onto the gel.

Remarks: When using FastDigest CseI for digestion, you need to copy at least two of its recognition sequences. FastDigest CseI may bind to severed DNA. This can cause DNA band drift during electrophoresis. To avoid atypical DNA electrophoresis band patterns, please use 6 loading dye & SDS solution< /a> (#R1151) Prepare the sample, or add SDS to the DNA digest and heat it before electrophoresis.

FastDigest Green Buffer provides the same high performance as colorless FastDigest Buffer in DNA digestion reactions and downstream applications. For applications requiring product analysis by fluorescence excitation (eg determination of concentration under UV light), the use of colorless FastDigest buffer is recommended.
