PureLink™ HiPure Plasmid Miniprep Kit is designed to efficiently isolate plasmid DNA from E. coli in 1.5 - 2 hours. The kit uses an anion exchange resin composed of small particles with uniform pore sizes to purify plasmid DNA to a level equivalent to two passes through a CsCl gradient (Figure 1). No additional steps are required to remove contaminants such as phenol, chloroform and CsCl, minimizing exposure and handling of hazardous materials.
High Yield and Efficient Endotoxin Removal
When cloning high copy number plasmids, the PureLink™ HiPure Plasmid Miniprep Kit can be used from 1-3 in about 1 hour. Purify up to 30µg of high-quality plasmid DNA from mL overnight E. coli cultures with endotoxin levels of 0.1-1 EU/µg (Table 1). Typically, A260/A280 > when diluting samples in Tris-HCl (pH 7.5) for DNA isolated using the PureLink™ HiPure Plasmid Miniprep Kit ; 1.80, indicating that the DNA does not contain proteins that may interfere with downstream applications.

Purified DNA is of ultrapure grade and is suitable for a variety of downstream applications, including those requiring higher purity DNA, e.g. Mammalian transfection (Figure 2), automated and manual DNA sequencing, PCR amplification, in vitro transcription, bacterial cell transformation, cloning and labeling.
