pBAD/TOPO™ ThioFusion™ Expression Kit is designed to achieve sulfur oxygenation in E. coli One-step cloning and regulated expression of fusion proteins (Figure 1). The pBAD/Thio-TOPO™ vector encodes His-Patch thioredoxin as the N-terminal fusion partner. Thioredoxin fusion can significantly increase the solubility of many difficult-to-express proteins and increase protein production. The vector includes the following additional features (Figure 2):

•araBAD promoter for tightly controlled expression in E. coli, topoisomerase I Activation Vector for 5-minute TOPO™ cloning of Taq-amplified PCR products
• C-terminal polyhistidine (6xHis) tag for purification with nickel chelating resin , and detected with anti-His (C-terminal) antibody
• C-terminal V5 epitope, for detection with anti-V5 antibody
• Enterokinase cleavage site for efficient cleavage with EKMax™ enterokinase N-terminal fusion tag
