Labeled calcium indicators are molecules that show an increase in fluorescence upon binding to Ca
2+. They can be used for a variety of calcium signaling studies, including measurement of Ca
2+ in cells and tissues with high levels of autofluorescence and Ca
2+ produced by photoreceptors and photoactivated chelators. sup> release detection. The AM ester form of this calcium indicator can be loaded onto the cells by adding the dissolved indicator directly to the culture dish containing the cultured cells. Fluorescence signals from these cells are typically detected using fluorescence microscopy.
Calcium Indicator (AM Ester) Specifications:
• Label (Excitation/Emission Wavelength of Ca2+– bound form): Rhod-2 (552/ 581 nm)
• Fluorescence intensity increases after binding Ca2+: >100 times
•Ca when there is no Mg2+ in the buffer Kd of 2+: ∼570 nM
• After binding to Ca2+, the fluorescence increases and the wavelength changes slightlyControl of heavy metal cations using TPEN In addition, BAPTA-based indicators can bind various heavy metal cations (such as Mn
2+, Zn
2 +, Pb
+2), the affinity is much higher than Ca
2+. Perturbations in calcium measurements due to the presence of these ions can be controlled using the heavy metal selective chelator
TPEN.
More Choices of Fluorescent Calcium Indicators We offer a large selection of Molecular Probes™ calcium indicators that can be used in a variety of experimental protocols, such as dextran forms to reduce leakage and zone Compartmentalized, BAPTA conjugates are used to detect high-amplitude calcium transients.
