FluxOR™ II Green Potassium Channel Detection Kit is a high-throughput screening (HTS) ) Next generation solutions for potassium channel and transporter activity. The FluxOR II Green Potassium Channel Assay Kit takes advantage of the permeability of potassium channels to thallium ions. It uses an improved thallium-sensitive fluorescent indicator dye, which can obtain a larger signal window and significantly improve the sensitivity of detection. In addition, the detection step is available in "cleaning" or "no cleaning" formats.
Features include:
• Higher detection sensitivity and larger signal window facilitate analysis of a wider range of potassium channels, including low expression or weakly conducting channels
• Compared to previous generation products 100% increased dynamic range compared to competitive products; >40% increased dynamic range compared to competitive products
• Homogeneous, no-wash solution reduces well-to-well variation and improves assay reproducibility
• Optimized for high-throughput screening applications
• Formats available for low to high-throughput users
Similar to previous generation assays, the FluxOR II Green Potassium Channel Assay exploits the potassium channel’s permeability to thallium sex. When a stimulus opens potassium channels, extracellular thallium flows into the cell along the concentration gradient. The ion channel or transporter activity is detected through a proprietary indicator dye preloaded into the cell. In the presence of thallium, the cell Solute fluorescence is enhanced. FluxOR II Detection The fluorescence observed in this manner is an indicator of ion channel activity or transport processes that allow thallium to enter the cell. This fluorescence signal quantitatively reflects the activity of thallium-permeable ion channels and transporters, including hERG, Kir2.1, and other pharmacologically important potassium channels. For the FluxOR II detection kit, an improved indicator dye was developed to obtain a larger signal window and improve detection sensitivity.
In addition to the modified fluorescent indicator dye, the FluxOR II Green Potassium Channel Detection Kit provides a concentrated thallium solution and necessary buffers for homogenization with greater target flexibility and ease of operation. form, which has been confirmed in cells stably expressing hERG. The detection step has two forms: "cleaning" or "no cleaning".
