Transferrin is a monomeric serum glycoprotein (∼80,000 daltons) that mediated through Endocytosis binds to specific receptors on the surface of vertebrate cells and delivers up to 2 Fe3+ atoms - labeled LDL complexes are a useful tool to study this phenomenon. After transferrin carrying iron enters the endosome, the acidic environment can promote the dissociation of iron ions from the transferrin-receptor complex. After iron is released, apotransferrin is recycled into the cell membrane, where it is released from the receptor to bind more iron. Therefore, fluorescent transferrin conjugates can be used with fluorescent LDL to differentiate between the lysote-directed pathway and the endosomal recycling pathway.
These experiments are typically performed by adding fluorescently labeled transferrin to cultured cells and analyzing them by microscopy. We offer biotinylated transferrin conjugates in over 10 fluorescent formats.
