The Thermo Scientific Cultured Cell Mitochondria Isolation Kit provides a versatile microcentrifuge tube method for isolation in approximately 40 minutes Isolation of intact mitochondria from cultured mammalian cell samples.
Features of the Cultured Cell Mitochondria Isolation Kit:
•
Fast—Isolates from 20 million cells in approximately 40 minutes (after cell harvest) Intact Mitochondria
•
Flexible—Process multiple samples simultaneously using reagent-based methods or use Doones homogenization for the highest possible yield
•
Available Optimization—Provides guidance for optimizing purity and yield using reagent-based methods
•
Benchtop-Easy—Separations performed in microcentrifuge tubes
•
Compatible< /b>—Isolated mitochondria can be lysed with detergents or processed for virtually any downstream application (including 2D electrophoresis)
Mitochondria isolation is often a tedious process that requires Doones homogenization method for single sample processing. The Mitochondria Isolation Kit uses a non-mechanical, reagent-based method to process multiple (6) cultured cell samples simultaneously in approximately 40 minutes. Cultured mammalian cell pellets are gently lysed using a proprietary formulation that yields the highest mitochondrial yields while minimizing damage to integrity.
The product description describes guidelines for optimizing purity and yield parameters. The kit also offers an optimized Dunes homogenization procedure that achieves up to two times higher mitochondrial recovery than reagent-based methods. Both methods separated mitochondrial and cytoplasmic fractions using different centrifugation speeds in a tabletop microcentrifuge and were completed in approximately 40 minutes (post-cell harvesting). Once isolated, mitochondria can be used for downstream applications (such as apoptosis, signal transduction, and metabolism studies) as well as to facilitate mitochondrial proteomics work.
