This gentle and easy-to-use kit isolates highly pure and functional mouse Treg cells from mouse lymph node or spleen samples . Your cells are not exposed to pressure conducted through the column. Isolated CD4+CD25+Foxp3+ Treg cells are magnetic bead-free and compatible with flow cytometry. Cells can be expanded very easily and retain their functional inhibitory capabilities. It is expected to obtain >90% purity grade Treg cells. More than 88% of isolated CD25+ cells express Foxp3. You'll save time and money and gain greater confidence in your results.

Method:
Negatively isolate CD4+ T cells, and then positively isolate CD25+ regulatory T cells. First, a cocktail of antibodies is added to the sample, and the antibodies will bind to non-CD4+ T cells. Mouse Depletion Dynabeads™ are then added, which will bind to the antibody-cell complexes within a short incubation time. Cells bound to magnetic beads were detached on a magnetic stand and discarded. The remaining unexposed mouse CD4+ T cells were then labeled with FlowComp™ CD25 Antibody and positive isolation was performed using FlowComp™ Dynabeads™. Isolated mouse regulatory T cells are released from Dynabeads™ after a short incubation with FlowComp™ Release Buffer. This isolation method is easy to use and scalable.
