Ham's F-12K (Kaighn's) Medium is an improved version of Ham's F-12 Nutrient Mixture. Ham's F-12K (Kaighn's) medium was developed specifically for primary human hepatocytes and selected rat and chicken hepatocytes in a serum-reduced environment.

Usage of Ham's F-12KHam's F-12K (Kaighn's) medium contains many ingredients that are not found in traditional basal medium, such as putrescine and thymidine , hypoxanthine, zinc, and higher levels of all amino acids and sodium pyruvate. With the addition of these components, only minimal amounts of serum or well-defined components need to be supplemented in the culture medium for certain cell types. Ham's F-12K (Kaighn's) medium does not contain protein or growth factors, so it is often supplemented with growth factors and
Fetal Bovine Serum (FBS). FBS concentration must be optimized for each cell line. Ham's F-12K (Kaighn's) medium uses a sodium bicarbonate buffer system (2.5 g/L) and therefore requires an environment of 5–10% CO
2 to maintain physiological pH.
