•Sulfo -NHS ester terminal is coupled with primary amine at pH 7-9 to form a stable amide bond
•Maleimide reacts with -SH group at pH 6.5-7.5 to form a stable thioether Connect
•Non-cleavable
•Water soluble (comparable to EMCS)
•Aliphatic spacers are less likely to elicit an immune response
