Whole blood gene expression profiling and profiling Becoming an increasingly important life science tool. Accurate measurement of blood-based molecular profiles of RNA transcripts is a critical need in research areas such as disease association studies, biomarker discovery and validation, and monitoring of drug regimens.

The highly unstable nature of in vitro RNA transcripts poses greater challenges to accurate expression analysis. In standard blood collection tubes, ongoing metabolic changes can dramatically affect gene expression profiles in as little as an hour (see figure). Therefore, the use of Applied Biosystems™ Tempus™ Blood RNA Tubes (SKU #4342792) is recommended. These tubes are designed to collect 3 mL of blood directly into plastic vacuum blood collection tubes containing RNA stabilizing reagent. RNA is stable in collection tubes at room temperature for up to five days and at 4°C for at least 7 days, so its expression profile is as shown above (shown in the figure).
Easy to perform
The Tempus™ system (Tempus™ Blood RNA Tubes and Tempus™ Centrifugal RNA Isolation Kit) has been tested for stable RNA expression profiles under a range of storage conditions capabilities were tested (as shown in the figure). This combination provides stable RNA, high yields of purified RNA (observed yields of 6–25 µg from normal healthy donor whole blood), and high quality RNA (A260/280 ratio greater than 1.9, RIN greater than 7.0) , suitable for downstream applications such as quantitative PCR, microarray profiling and other RNA analysis methods.
