Dynabeads™ M-270 Streptavidin is a biotinylated nucleic acid, antibody or other biotinylated ligand and target The gold standard for molecular separation and processing. The extremely high binding affinity of the streptavidin-biotin interaction (Kd=10-15) has been used in a large number of applications. Advantages and Features:
• Direct and rapid separation of any biotinylated molecule
• Flexible protocol with mild and efficient liquid phase reaction kinetics
• Very low non-specific nucleotide and nucleic acid binding
• Low aggregation in high-salt hybridization buffers
• Ideal for nucleic acid applications with extreme requirements
• Low levels of non-specific binding to small, negatively charged proteins
• Used in medically compliant materials Devices are manufactured using a validated cGMP manufacturing process
• Ideal for automation solutions
Information about Dynabeads™ M-270 Streptavidin
These uniformly sized Superparamagnetic microbeads are 2.8 µm in diameter and have a single layer of covalently coupled recombinant streptavidin on their surface rather than multiple layers. This design leaves the vast majority of biotin-binding sites available for binding not only free biotin but also biotinylated ligands/targets. They are hydrophilic, negatively charged, and exhibit fast liquid phase reaction kinetics. Its specific, defined surface enables efficient capture, separation and downstream processing. The single layer of streptavidin ensures negligible leakage levels without excess adsorbed streptavidin, ensuring consistent and reproducible results. Applications
Streptavidin-conjugated Dynabeads™ have been used and referenced in a variety of applications over the past 15 years. For example, direct/indirect separation and downstream operations of nucleic acids, proteins/peptides, and other target molecules. Suitable for sequence-specific DNA/RNA capture in nucleic acid-based diagnostics, especially samples with high chaotropic salt concentrations, immunoassays involving biotinylated small antigens, and applications incompatible with BSA (beads are not blocked with BSA). Easily adapt to automated processes. Dynabeads™ are used in more than 25,000 routine IVD instruments worldwide. The products have high standards in terms of reproducibility (intra- and inter-batch) and automated operation, providing reliability of results.
Binding Capacity
The size of the molecule and the biotinylation step will affect binding capacity. Binding capacity also depends on available space and charge interactions between beads and molecules and between molecules. After binding, each avidin molecule on the bead surface has two or three biotin binding sites. 1 mg Dynabeads™ M-270 Streptavidin typically binds:
•>950 pmole free biotin
• ∼200 pmol biotinylated peptide
• ∼10 µg biotin IgG
• ∼10 µg ds-DNA
• ∼200 pmol single-stranded oligonucleotide
