Pierce High Capacity Endotoxin Removal Resin combines porous cellulose beads with an FDA-approved food preservative, poly(epsilon-lysine), to selectively bind endotoxins affinity ligand. Use our centrifuge cup formats to reduce endotoxin levels in protein samples by ≥99% and achieve ≥85% protein recovery in as little as 1 hour. Modified polylysine affinity ligands avoid toxicity issues associated with alternative technologies using polymyxin B ligands and sodium deoxycholate buffer.

The presence of endotoxins in biologically derived products prepared for research, vaccine or therapeutic use is a major concern because of the diverse and potentially harmful biological responses to these molecules. Endotoxin contamination of proteins purified from bacterial cultures is caused by residual lipopolysaccharide (LPS) derived from the outer cell membrane of Gram-negative bacteria such as Escherichia coli. Protein samples contaminated with endotoxins can be transfected into cells or injected into animal hosts, which can initiate a strong immune response and produce systemic inflammatory response syndrome (SIRS) and/or sepsis. Traditional endotoxin removal resins using polymyxin B and sodium deoxycholate to remove endotoxins from samples also have the negative effects of neurotoxicity and acute tubular necrosis.

Pierce High Capacity Endotoxin Removal Resin is a porous cellulose bead whose surface is modified with covalently linked polylysine chains that have high affinity for LPS but lack other endotoxin-binding molecules toxicity. Utilizing a binding capacity of 2,000,000 EU/mL, endotoxin levels can be reduced by 99% in samples with an initial endotoxin level of 10,000 EU/mL. Typical protein samples treated with high-capacity endotoxin removal resin have final endotoxin concentrations below 5 EU/mL. This resin is available in slurry format for custom packed endotoxin removal columns suitable for gravity flow or flow rates in the range 10-15 mL/h.
