Pierce streptavidin magnetic beads use a recombinant streptavidin with a mass of 53kDa and an isoelectric point (pI) close to neutral. The protein is a tetramer with four biotin-binding sites. Unlike avidin, streptavidin does not contain sugar groups and therefore has very low non-specific binding. The high-affinity binding between streptavidin and biotin will only be destroyed under very harsh conditions, such as boiling in SDS-PAGE loading buffer or 8M guanidine·HCl at pH 1.5. . Therefore, typically the biotinylated bait protein will not be eluted at the same time as the capture protein in the interacting complex.
