ViraPower™ HiPerform™ T-REx™ Gateway™ Vector Kit contains Gateway™-adjusted ViraPower™ HiPerform™ tetracycline-adjusted slow The viral expression vector pLenti6.3⁄TO⁄V5-DEST™ is used for simple recombination-based cloning and lentiviral-based, regulated, high-level expression of target genes in dividing and non-dividing mammalian cells. The pLenti6.3⁄ TO⁄ V5-DEST™ vector is equipped with two key genetic elements that make it a HiPerform™ vector: the woodchuck post-transcriptional regulatory element (WPRE) and the central polypurine pipeline (cPPT) from the HIV-1 integrase gene ) sequences used to produce at least a 4-fold increase in protein expression relative to vectors lacking these elements.
Advantages
•Replication-incompetent lentivirus can be generated for transduction of dividing and non-dividing mammalian cells
•Gateway™ technology can be used to perform Recombinant simple cloning
•Stable, long-term, tetracycline-regulated expression
•Enhanced protein expression up to or more than 4 times (relative to traditional lentiviral expression system)
Main Features
• Features WPRE from woodchuck hepatitis virus to increase transgene expression and cPPT from the HIV-1 integrase gene to increase copy number of the lentivirus integrated into the host genome, thereby increasing viral titers . Compared to other vectors without these elements, WPRE and cPPT increased protein expression at least fourfold in most cell types.
• Features a hybrid promoter consisting of the human cytomegalovirus (CMV) promoter and two tetracycline operator 2 (TetO2) sites for high-level, regulated expression of target genes
• Features rice-killing Blasticidin selection marker for stable selection under the control of the SV40 promoter
Kit includes
• pLenti6.3⁄TO⁄V5-DEST™ vector
• pLenti3.3⁄TR Vector
• pLenti6.3⁄TO⁄V5-GW⁄lacZ Plasmid
• Tetracycline
• One Shot™ Stbl3™ Chemically Competent E. coli (Cat. No. C737303)
• pENTRTM Gus positive control plasmid.
