Hoechst 33342 is a commonly used cell-permeable nuclear counterstain that emits blue fluorescence after binding to DNA. . With NucBlue™ Live ReadyProbes™ reagents, we've formulated a room-temperature stable solution of this classic stain in a convenient dropper bottle. Just two drops per milliliter are enough to stain cells.

Cell imaging applicationsHoechst 33342 (2'-[4-ethoxyphenyl]-5-[4-methyl-1-piperazinyl]-2 ,5'-di-1H-benzimidazole), including a large Stokes shift, making it ideally suited for use with green (Alexa Fluor™ 488, FITC, GFP) and red (Alexa Fluor™ 594, Texas Red™, rhodamine, mCherry, mKate-2) fluorophores are used together for multicolor experiments. Due to its high affinity for DNA, Hoechst 33342 is also commonly used to identify condensed nuclei of apoptotic cells in cell counting, cell cycle and cell replication studies. It is used in conjunction with Click-iT™ EdU or BrdU staining for cell cycle studies as a high-content Nucleus segmentation tools in imaging analysis can also sort cells based on their DNA content.
Usage RecommendationsNucBlue™ Live Cell Stain can be added directly to cells in complete medium or buffered solutions.
• In most cases, 2 drops/mL and an incubation time of 15 to 30 minutes will produce bright nuclear staining; however, optimization may be required for some cell types, conditions, and applications. In this case, simply increase or decrease the number of drops until you obtain the optimal stain intensity. In most cases, if cells are not washed before imaging, staining intensity increases with time.
•After binding to DNA, NucBlue™ live cell stain can be excited by UV light at 360 nm, with a maximum emission wavelength of 460 nm. Detection can be performed with blue/cyan filters such as DAPI filters, blue GFP filters, or the Semrock BrightLine™ Alexa Fluor™ 350 Dye Filter Kit.
