MagMAX™ FFPE DNA/RNA Ultra Kit is designed for use in formaldehyde- or paraformaldehyde-fixed paraffin-embedded (FFPE ) Continuous isolation of DNA and RNA from tissue samples. DNA and RNA are recovered in separate eluates, both compatible with a variety of applications including real-time PCR and next-generation sequencing. Isolating RNA and DNA from the same FFPE sample makes this kit Oncomine™ Comprehensive and Ideal sample preparation method for Oncomine™ Focus assays.

Unlocking the information contained in FFPE samples is critical for cancer research. Common nucleic acid isolation methods require you to choose which nucleic acids to recover from a given sample, while losing others (and any information their analysis may yield). Alternatively, co-purify DNA and RNA, or split the sample into two equal parts so that each part can be purified.

Get more information with less sample input
The MagMAX FFPE DNA/RNA Ultra kit protocol works by first isolating DNA from the FFPE sample and then removing the DNA from the sample. Recovery of RNA from supernatant solution solves these problems. Nucleic acids are prepared as separate eluates and then prepared for downstream analysis. This enables more comprehensive analysis of precious samples, including the study of important biomarkers such as hotspot mutations, CNVs, gene fusions and indels.

MagMAX Magnetic Bead Method – Advantages of Purification
The kit’s – Magnetic Bead Method Purification specifications can be easily increased or decreased using a magnetic stand for manual processing or using a magnetic particle processor such as Number of samples processed by KingFisher™ Duo Prime or KingFisher™ Flex Processor).

Magnetic beads offer many advantages over other technologies for isolating nucleic acids. Magnetic beads bind RNA and DNA more efficiently than glass fiber filter columns, resulting in higher and more consistent yields. Additionally, since filter columns and vacuum manifolds are not used, there is no risk of undigested tissue particles clogging these components during the extraction process. This clogging problem is particularly concerning for fibrous FFPE tissue samples that were not completely digested during proteinase K treatment.

Optimized bead surface chemistry
The Dynabeads MyOne Silane used in the MagMAX FFPE DNA/RNA Ultra Kit has an optimized silica-like surface chemistry and high specificity surface area for efficient kinetics and highly sensitive nucleic acid capture. These properties make Dynabeads MyOne Silane ideal for recovering RNA and DNA targets found in FFPE samples. After capture, the two nucleic acids are eluted separately in a volume of 50 μL and ready for downstream applications.
