Thermo Scientific Pierce Anti-DYKDDDDK Affinity Resin is a polymer for in vitro protein expression systems, bacteria, yeast and mammalian cells. A fast and convenient method for purification and immunoprecipitation (IP) of DYKDDDDK-tagged proteins expressed in . The amino acid sequence DYKDDDDK (commonly known as FLAG) is recognized by a high-affinity rat monoclonal antibody (clone L5) covalently linked to UltraLink Biosupport. UltraLink Biosupport is a rigid, hydrophilic, highly cross-linked, copolymeric and porous resin with high coupling capacity. The resin's porosity, stiffness, and durability make it suitable for medium-pressure, fast-flow techniques involving large sample volumes.
For protein purification or IP, add resin to samples containing DYKDDDDK-tagged proteins, where the tag is located at the N or C terminus. After capturing the DYKDDDDK-tagged protein, non-specific binding proteins are washed away, and the bound target protein is separated with an elution buffer. Pierce Anti-DYKDDDDK Affinity Resin is recommended for use with spin columns or purification columns in FPLC instruments.
Features include:
•Specificity — Unique base beads and highly specific antibodies minimize off-target binding (low non-specific binding)
•High Purity — Optimized bind-wash-elute protocol enables high purity
•High Yield — Special antibody conjugation method enables high yield
•Fast — The entire purification protocol typically takes less than 40 minutes
•Economic — The purification protocol can be reused multiple times
• Versatile—can be used for centrifugation, gravity, or FPLC-based purification and enrichment methods
•Scalable—available in 1 mL, 5 mL, and 50 mL precipitation resin packaging For smaller scale immunoprecipitation or larger scale purification
