Thermo Scientific Pierce BS3 (Sulfo-DSS) is a bis(sulfosuccinimide Amine) suberate, a homobifunctional, water-soluble, non-cleavable and membrane-impermeable amine-amine cross-linker.

BS3 An amine-reactive N-hydroxysulfosuccinimide (NHS) at each end of the 8-carbon spacer arm ester. NHS esters react with primary amines at pH 7-9 to form stable amide bonds and release the N-hydroxysulfosuccinimide leaving group. Proteins, including antibodies, often have several primary amines in the side chains of lysine (K) residues and at the N-terminus of each polypeptide, and these primary amines can serve as targets for NHS ester cross-linking reagents.

No-Weigh Microtube Processing:
Eight microtubes each contain 2 mg of cross-linker. Pierce the aluminum foil with a pipette tip and add DMSO or DMF to DSS or water buffer to BS3. Store microtube drain plugs in the aluminum foil bags provided. Used microtubes can be cut off from unused microtubes and discarded.

Because it contains hydrophilic sulfonyl groups, BS3 The cross-linker is soluble in water and many common buffers up to ∼100 mM, thus avoiding the use of organic solvents that may disrupt protein structure. The water-soluble analogue of BS3, DSS, is also suitable for applications requiring a less hydrophilic cross-linker (e.g. to produce intracellular cross-links). DSS and BS3 have essentially the same cross-linking activity for primary amines.
