Dual probe detection suitable for multiple platforms
Like our popular LIVE/DEAD® cell viability/cytotoxicity Detection, the new LIVE/DEAD® Cell Imaging Kit is based on cell-permeable dyes suitable for staining live cells, and non-cell-permeable dyes suitable for dead and dying cells (characterized by damaged cell membranes). To make this important assay applicable to a variety of imaging platforms, the components of the LIVE/DEAD® Cell Imaging Kit are optimized for use with common green and red imaging filters such as FITC and Texas Red. The live cell component produces strong and uniform green fluorescence (excitation/emission wavelengths of 488 nm/515 nm) in living cells. The dead cell fraction produces nuclear red fluorescence (excitation/emission wavelength of 570 nm/602 nm) mainly in cells with damaged cell membranes as a strong indication of cell death and cytotoxicity (Figure 1).

Quickly and accurately measure cell viability and cytotoxicity
LIVE/DEAD® Cell Imaging Kit can more sensitively detect key indicators of cell health: cell viability, living cells /dead cell ratio and cytotoxicity. The components of the LIVE/DEAD® Cell Imaging Kit are formulated for ease of use and require only low dilutions.

Detection Principle
Using the LIVE/DEAD® Cell Imaging Kit, the almost non-fluorescent cell-permeable calcein AM and Enzymatic conversion of high-intensity fluorescent calcein identifies ubiquitous intracellular esterase activity, thereby distinguishing living cells. The red component of the LIVE/DEAD® Cell Imaging Kit is cell impermeable and therefore can only enter cells with damaged cell membranes. Upon binding to DNA in dying and dying cells, it produces bright red fluorescence. The detection technique itself has low background fluorescence levels because the dye barely fluoresces until it interacts with cells. The LIVE/DEAD® Cell Imaging Kit was selected based on the brightness, spectral properties (FITC and Texas Red filters) and ease of use of the fluorophores included. Because it is packaged for workflow convenience, live and dead cell fractions in cell populations can be determined easily and quickly.
