Thermo Scientific Pierce DSS (also known as disuccinimidyl suberate) is a non-cleavable membrane-penetrable Sexual cross-linker with an amine-reactive N-hydroxysuccinimide (NHS) ester at each terminus of the 8-carbon spacer arm.

NHS esters react with primary amines at pH 7-9 to form stable amide bonds and simultaneously release the N-hydroxysuccinimide leaving group. Proteins, including antibodies, often have several primary amines in the side chains of lysine (K) residues and at the N-terminus of each polypeptide, and these primary amines can serve as targets for NHS ester cross-linking reagents. DSS is first dissolved in an organic solvent (such as DMF or DMSO) and then added to the aqueous cross-linking reaction. The water-soluble analogue of DSS, BS3, is also suitable for applications requiring hydrophilic cross-linkers (e.g. to produce cell surface cross-links). The cross-linking activity of DSS and BS3 towards primary amines is essentially the same.
