Thermo Scientific HisPur cobalt resin is a tetravalent cobalt resin containing divalent cobalt (Co2+). Ligand-chelating agarose resin is used to obtain high-purity His-tagged proteins without metal contamination.

HisPur cobalt resin can provide higher Binds histidine-tagged proteins specifically (lower off-target binding) and releases them using milder conditions (lower concentrations of imidazole). Although Ni(2+) chelating resins provide higher protein yields, they undergo a certain degree of indiscriminate binding, resulting in poor purity. Often, additional purification steps are required. In contrast, cobalt maximizes protein purity without sacrificing protein yield. Compared to nickel resin, HisPur cobalt resin binds fewer non-specific proteins, exhibits lower metal leaching rates and enables more relaxed elution conditions.

HisPur Cobalt IMAC Resin specifically binds His-tagged recombinant proteins that can be efficiently recovered by mild imidazole elution. More than 10 mg of His-tagged protein was bound per ml of resin using a 28 kDa fusion protein expressed and purified from E. coli lysate.

A variety of His-tagged proteins can be recovered with purity similar to or higher than that obtained from other available nickel or cobalt IMAC resins using either centrifugation or gravity flow column formats. The purification performance of HisPur Cobalt Resin was compared with IMAC resins from other suppliers using a variety of His-tagged proteins of varying sizes and expression levels. Results show that HisPur cobalt resin can achieve higher protein purity without sacrificing protein yield.
