Use modified nucleotide ethynyluridine (EU, E10345) or use Click-iT nascent RNA capture reagent Cassette (C-10365) detects temporal and spatial changes in total RNA transcription in vitro and in vivo.
Both detection methods use powerful click chemistry, a simple and reliable two-step technology. In the first step, the cell or animal is fed with an alkyne-containing biomolecule and actively incorporated into the RNA. The assay utilizes a chemoselective linkage or "click" reaction between an azide and an alkyne, using the corresponding azide-containing dye or hapten with the Click-iT™ Cell Reaction Buffer Kit to detect modified proteins . Cellular analysis can be performed using fluorescence microscopy, flow cytometry, or high-content imaging and analysis (HCS).
