Ambion™ ULTRAhyb™-Oligo uses a unique formula to be used with oligonucleotide probes for Northern analysis. It contains a unique blend of hybridization promoters and blocking reagents that greatly increases hybridization levels so that signals that once took days to observe can now be visualized in just hours. Supplied in a 125 mL bottle.

Using standard hybridization buffers, only 1–5% of the target molecules on the blot hybridize to the probe, making blotting a relatively insensitive method of nucleic acid analysis ( Vernier et al. (1996) Anal Biochem 235:11-19). Hybridization reactions are performed using ULTRAhyb™ ultra-sensitive hybridization buffer, allowing detection of as few as 10,000 molecules. Oligonucleotides of length 26-45 are used with ULTRAhyb™-Oligo. Ideal for use with oligonucleotides with T_m values of 78°–90°C and 47–62% GC content. However, the optimal hybridization temperature for oligonucleotide probes with different GC contents and T_m values should be determined empirically.
