Invitrogen™ CellBlox™ blocking buffer is formulated to block non-specific binding of Invitrogen™ NovaFluor™ tags to cells . These nonspecific interactions can result in higher background labeling. CellBlox Blocking Buffer is a non-antibody, non-protein-blocking solution that should be used every time NovaFluor dyes are used to label any cell type to minimize background labeling. (Figure 1)
CellBlox Blocking Buffer is also recommended for use with cyanine-containing dyes or cyanine-containing tandem dyes to block non-specific interactions with monocytes and macrophages, thereby minimizing Reduce background marks. (Figure 2)
When using CellBlox Blocking Buffer, only minimal changes need to be made to most flow cytometry staining protocols. Before adding antibodies, add 5µL of CellBlox Blocking Buffer directly to the solution containing 103 - 108 cells in cell suspension, final chromosome volume is 100 µL. CellBlox Blocking Buffer can be added to the antibody mixture before labeling cells by adding 5 µL of CellBlox Blocking Buffer to each stained sample to be labeled with the antibody mixture for a final chromosome volume of 100 µL
• Labeling When using cells, be sure to use CellBlox Blocking Buffer with NovaFluor dyes for excellent background subtraction.
•CellBlox Blocking Buffer is compatible with all fluorophores and Invitrogen™ LIVE/DEAD™ fixable dead cell stains.
•CellBlox Blocking Buffer can be used with any fluorophore-antibody conjugate as a high-performance monocyte and macrophage blocking solution.
•CellBlox Blocking Buffer is compatible with other blocking reagents such as Fc Block, Blocking Protein, Brilliant Staining Buffer and Super Bright Complete Staining Buffer.
•Labeled Antibodies - CellBlox blocking buffer is not required when capturing beads
