Click-iT™ Tetramethylrhodamine (TAMRA) Glycoprotein Assay Kit provides The second part of a simple and reliable two-step technique for the identification and characterization of glycoproteins by one- or two-dimensional gel electrophoresis. In the second step, the azide handle is incorporated into the proteoglycan structure through chemoselective linkages between azides and alkynes using the Click-iT™ Metabolic Labeling Reagent or the Click-iT™ Enzymatic Labeling System. or click reaction to detect azide-modified glycoproteins. Gels containing TAMRA-tagged glycoproteins were then stained using Multiplexed Proteomics™ technology, SYPRO™ Ruby Total Protein Stain, and PRO-Q™ Emerald Glycoprotein Stain to analyze glycoprotein subclasses, total proteins, and and differential analysis of total glycoproteins. Click-iT™ modified glycoproteins are also compatible with downstream LC-MS/MS and MALDI MS analysis for further identification.
