2DCFDA is the chloromethane of H
2DCFDA derivatives that can be used as indicators of reactive oxygen species (ROS) in cells. The retention rate of this indicator in living cells is much higher than that of H
2DCFDA. CM-H
2DCFDA can passively diffuse into cells, its acetate group is cleaved by intracellular esterase, and its thiol-reactive chloromethyl group interacts with intracellular glutathione and Other thiols react. Subsequent oxidation produces a fluorescent adduct that remains within the cell and therefore facilitates long-term studies.
ROS indicator quality standards:
•Excitation/emission wavelength: ∼492–495/517–527 nm
• Product is air sensitive and should be stored in dry argon or In nitrogen
• The product can be dissolved in DMSO, DMF or ethanol for use
• The indicator is permeable to cells (see the literature for cell loading protocols)
• Flow cytometer, fluorometer, micropore can be used Monitor the fluorescence signal with a plate reader or fluorescence microscope (equipped with an excitation light source and filters suitable for fluorescein)
