Thermo Scientific Phusion U Multipass PCR Master Mix is a ready-to-use 2X end-point PCR master mix designed for use in a single Amplify multiple targets simultaneously in a tube. More than 20 primer pairs can be added simultaneously to a reaction system to achieve extremely high specificity and efficient multi-pathway analysis over a wide range of primer and template concentrations.

Phusion U Multiplex PCR Master Mix contains a proprietary buffer that adds balanced concentrations of all PCR components, eliminating the need for tedious and time-consuming optimization of reaction conditions. Phusion U Green Multipass PCR Master Mix (F-564S or F-564L) further simplifies workflow - it contains a density reagent and two electrophoretic tracking dyes for direct loading of PCR products onto the gel.

This master mix uses Phusion U Hot Start DNA Polymerase, a high-performance enzyme developed using fusion technology. Similar to other enzymes in the Phusion family, Phusion U DNA Polymerase introduces more nucleotides per binding. This significantly reduces the time of the extension step, but results in a higher yield of PCR product, thus reducing the time of the overall experimental workflow. Significantly improved processivity also enables successful multiplex PCR of difficult targets, such as templates with high GC content or templates of poor purity. Phusion U Multiplex PCR Master Mix is highly tolerant to a wide range of PCR inhibitors and can even be used for amplification with unpurified samples such as blood or serum.

The specificity of multiplex PCR is improved by the Affibody ligand-mediated hot start mechanism. Reversibly binding Affibody ligands inhibit polymerase activity at room temperature, thereby preventing amplification of non-specific products and formation of primer-dimers.
