FUN™ 1 is a unique two-color fluorescent viability probe for yeasts and fungi Needle. FUN™ 1 stain passively diffuses into a variety of cell types and provides initial staining of the cytoplasm with diffuse green fluorescence. However, in some common yeast and fungal species, subsequent processing of the dye by living cells results in the formation of apparently compact vacuolar structures that display significant red fluorescence accompanied by a reduction in green cytoplasmic fluorescence. The formation of vacuolar structures requires the integrity of the cytoplasmic membrane and the metabolic capacity of the cell. The fluorescence of dead cells is bright yellow-green and has no obvious red structure.
