After the dye binds to the small grooves of DNA, it emits a distinct fluorescence spectrum, and the fluorescence intensity depends on the dye:base ratio. Hoechst 33342 Ready Flow ready-to-use dye is excited by UV light and has an emission peak at 460 nm.
