All components in the kit contain premixed reagents, reducing pipetting steps and providing convenience. The End Repair Enzyme Blend contains optimized T4 DNA Polymerase and Klenow Fragment Blend for efficient blunt-ending of fragmented DNA, and T4 Polynucleotide Kinase (PNK) for efficient phosphorylation of DNA ends. 10X End Repair Reaction Mix contains optimized reaction buffer, ATP, and dNTPs.

Samples such as fragmented genomic DNA (restriction enzyme digestion, nebulization or sonication), restriction enzyme digestion plasmid DNA, double-stranded cDNA, and PCR products containing dA overhangs are all compatible with This kit is compatible.

Features

In the DNA end repair reaction, fragmented DNA is converted into blunt-ended DNA containing 5'-phosphate groups and 3'-hydroxyl groups. The 5'→3' polymerase activity of the end repair enzyme mixture fills in the 5' overhanging DNA ends, while the 3'→→5' exonuclease activity removes the 3'-overhanging ends. T4 PNK adds 5'-phosphate to the ends of non-phosphorylated DNA fragments such as PCR products. (SeeTable 1 and Figure 1 below).
