Our Flp-In™ complete system allows the integration and expression of your target genes at specific genomic locations in mammalian cells . The Flp-In system involves the introduction of Flp recombination target (FRT) sites into the genome of selected mammalian cell lines. Then, the expression vector containing your target gene is integrated into the genome through DNA recombination mediated by Flp recombinase at the FRT site.
The main components of the Flp-In™ System include:
•An Flp-In. target site vector, pFRT⁄lacZeo, for generating host cell lines containing integrated FRT sites
• Expression plasmid containing an FRT site linked to a hygromycin resistance gene for use in stable cell lines expressing the gene of interest under the control of the human cytomegalovirus (CMV) immediate early enhancer⁄ promoter Flp recombinase-mediated integration and selection
• Flp recombinase expression plasmid pOG44 for Flp recombinase expression under the control of the human CMV promoter
• Contains the chloramphenicol acetyltransferase (CAT) gene The control expression plasmid, when co-transfected with pOG44 into the Flp-In. host cell line, expresses CAT
