AccurateSuccessful cell cycle analysis requires dyes that are DNA-selective and stain cells in a uniform pattern with minimal fluorescence variability. Vybrant™ DyeCycle™ Green stain is ideal for analyzing DNA content in living cells because the stain is cell permeable and, upon binding to double-stranded DNA, emits a fluorescent signal proportional to the DNA mass (see figure).

Simple, reliable staining protocolFor cell analysis, prepare only flow cytometry tubes containing 1 mL of cells at 1 × 10
6 cells/mL. Add 2 µL of Vybrant™ DyeCycle™ Green stain to each tube. The final stain concentration is 10 µM. Incubate for 30 minutes at 37°C in the dark. Analyze samples in a flow cytometer using 488 nm excitation and green emission wavelengths.
