Thermo Scientific EZ-Link NHS-PEG4-Biotin is a pegylated water-soluble reagent for antibodies, Simple and efficient biotin labeling of proteins and other primary amine-containing macromolecules.
EZ-Link NHS-PEG4-Biotin Features:
•
Protein Labeling—uses streptavidin probes or resins to label antibodies or other Biotinylation of proteins for detection or purification
•
Amine reactivity—reacts with primary amines (-NH2), such as the side chain of lysine (K) or the amino group of a polypeptide Terminal
•
PEGylated–spacer arm contains a hydrophilic 4-unit polyethylene glycol (PEG) group
•
Enhanced solubility–Poly Glycolation makes the biotinylated molecule water-soluble and helps prevent aggregation of biotinylated antibodies stored in solution
•
Irreversible—forms permanent amide bonds; the spacer arm cannot be Cleavage
•
Long Distance – The spacer arm (total length added to the target) is 29 Angstroms; this reduces steric hindrance to binding to the avidin molecule
NHS- PEG4 Biotin is a long (29.0Å) pegylated, water-soluble, NHS ester biotinylation reagent used to label amines and maximize the solubility of antibodies and other proteins. The
N-hydroxysuccinimide (NHS) group reacts specifically and efficiently with lysine and the N-terminal amino group to form a stable amide bond. Hydrophilicity
Polyethylene glycol (PEG) The spacer arm imparts water solubility, transferring it to the biotinylated molecule, thereby reducing storage in solution aggregation of labeled proteins. The PEG spacer arm also provides a long, flexible linkage to the reagent to minimize steric hindrance when binding to the avidin molecule.
We manufacture biotin reagents to ensure the highest possible level of overall product integrity, consistency and performance for intended research applications.
N-hydroxysulfosuccinimide (NHS) ester of biotin is a biotinylation reagent Widely used type. NHS-activated biotin reacts efficiently with primary amine groups (-NH2) in alkaline buffers to form stable amide bonds. Proteins (such as antibodies) often have several primary amines that can be targeted for labeling, including the side chain of a lysine (K) residue and the N-terminus of each polypeptide.
BiotinThe various NHS ester reagents vary in length, solubility, cell permeability and cleavability. Non-sulfonated NHS-Biotin is cell permeable but must be dissolved in organic solvents such as DMSO or DMF. Sulfo-NHS biotin (and biotin with a PEGylated spacer) can be dissolved directly in water but is not membrane permeable. Various types containing disulfide bonds can be cleaved using reducing agents, thereby separating the biotin groups from the labeled protein.
