Dynabeads M-280 Streptavidin is used to isolate and process biotinylated nucleic acids, antibodies or other biotinylated ligands and the gold standard for targets. The extremely high binding affinity of the streptavidin-biotin interaction (Kd=10-15) has been used in a large number of applications. Advantages and characteristics:
•Can directly and quickly separate any biotinylated molecule
•Low-charged and neutral beads, ideal for binding DNA fragments, proteins, peptides and antibodies
•Flexible solutions with Mild and efficient liquid-phase reaction kinetics
• Biomagnetic experiment protocol can be easily and flexibly adjusted for the automation platform
• High batch-to-batch reproducibility, ensuring the consistency of results in applications
Information about Dynabeads M-280 Streptavidin
These uniform superparamagnetic beads are 2.8 µm in diameter and feature a single layer rather than multiple layers of recombinant streptavidin, covalent Coupled to the surface and further blocked with BSA. The monolayer of streptavidin leaves the vast majority of biotin binding sites available for binding not only free biotin but also biotinylated ligands/targets. It exhibits fast liquid phase reaction kinetics. Its specific, defined surface enables efficient capture, separation and downstream processing. The single layer of streptavidin ensures negligible leakage levels without excess adsorbed streptavidin, ensuring consistent and reproducible results. Applications
Dynabeads M-280 Streptavidin has been used and referenced in many applications over the past 15 years. Key applications include using the Illumina platform to prepare single-stranded DNA templates to prepare 2–5 kb samples for paired library sequencing, isolating RNA and DNA-binding proteins, immobilizing large DNA fragments, purifying sequencing products, and specifically capturing nucleic acids. Dynabeads are used in more than 25,000 conventional IVD machines worldwide. The products have high standards in terms of reproducibility (intra- and inter-batch) and automated operation, providing reliability of results.
Binding Capacity
The size of the molecule and the biotinylation step will affect binding capacity. Binding capacity also depends on available space and charge interactions between beads and molecules and between molecules. After binding, each avidin molecule on the bead surface has two or three biotin binding sites. 1 mg Dynabeads M-280 Streptavidin typically binds:
•650–900 pmole free biotin
• ∼200 pmol biotinylated peptide
• ∼10 µg biotinylated IgG
• ∼10 µg ds-DNA
• ∼200 pmol single-stranded oligonucleotide
