Pierce Detergent Compatible Bradford Assay Kit is an adaptation of the well-known Bradford Coomassie dye for total protein quantitation Quick, ready-to-use improvements combined with colorimetry. Bradford's reagent's unique additives make it compatible with detergents and lysis reagents commonly used in life science research at concentrations of 1% or higher (including Triton X-100 and NP-40).

Similar to the Bradford method, the Coomassie dye binds to the protein in acidic medium, resulting in an immediate change in the absorption maximum from 465 nm to 595 nm, with accompanying color From green to blue. Furthermore, the assay only takes 10 minutes to complete.

Protein assays can be performed in test tube or microplate format. Standard working range is 100 – 1500 μ-g/mL with 1% detergent (or higher in some cases). Protein concentration is estimated by reference to the absorbance obtained against a series of dilutions of a standard protein, usually bovine serum albumin (BSA), which is measured together with an unknown sample. Because the color response of Coomassie dyes is nonlinear with increasing protein concentration, a standard curve must be completed with each assay. Standards can be used directly and do not need to be prepared in the same detergent found in the test samples.
