Thermo Scientific EZ-Link Alkoxyamine-PEG12-Biotin is a biotinylation reagent that contains a A 12-unit polyethylene glycol (PEG) spacer suitable for biotinylation of macromolecules in carbohydrate groups that have been oxidized to form aldehydes.

EZ-Link Alkoxyamine-PEG12-Biotin Features:

Glycoprotein Labeling—Biotinylation of glycosylated proteins (sialic acid residues) for detection or purification using streptavidin probes or resins
Cell surface markers—Biotinylate and isolate cell surface glycoproteins; reagents are not permeable Whole cell membrane
Aldehyde reactivity—reacts with aldehydes formed by glycosyl periodate oxidation
Alkoxyamine activation—amino oxygen The bond formed by the hydroxyl group is more stable than the bond formed by the hydrazide reagent
PEGylation—the spacer arm contains a hydrophilic 12-unit polyethylene glycol (PEG) group< br>•Enhanced solubility—PEGylation makes biotinylated molecules water-soluble, helping to prevent
irreversible aggregation of biotinylated antibodies stored in solution< /b>—Forms stable (permanent) oxime bonds; spacer arms cannot be cleaved
Solubility—Often dissolved in DMSO to prepare concentrated stock solutions
Long distance< /b>—The spacer arm (total length added to the target) is 55.4 Angstroms

Aminoxy-biotin reagents such as this one are useful for targeting glycoproteins and others with oxidizable polysaccharide groups Molecules are biotinylated. An alkoxyamine group (also called an aminooxy or aminooxy group) is coupled to an aldehyde that oxidizes a sugar. EZ-Link Alkoxyamine-PEG-Biotin Reagent contains a versatile extended spacer arm that is flexible, non-immunogenic, hydrophilicpoly Ethylene glycol (PEG), which imparts water solubility to the labeling molecule. Therefore, antibodies labeled with a pegylated biotin reagent show less aggregation when stored in solution than antibodies labeled with only a hydrocarbon spacer reagent.

We manufacture biotin reagents to ensure the highest possible level of overall product integrity, consistency and performance for intended research applications.

Biotinylation reagents vary in reactivity, length, solubility, cell permeability, and cleavability. Hydrazides and alkoxyamines are two types oftype of carbonyl reactive group. Alkoxyamines (—O-NH2) react specifically with aldehyde groups under near-neutral conditions to form stable oxime bonds. The reaction is more efficient in the presence of aniline (Product No. 88944). Alternatively, you can use EDC carbodiimide chemistryCouples alkoxyamines to carboxylic acids.

Sodium periodate (Product No. 20504) can be used to pass the sugar glycol group Partial oxidation generates reactive aldehyde groups in glycoproteins and other polysaccharide compounds. Sialic acid residues are common components of protein glycosylation and are easily converted to aldehydes with 1 mM NaIO4.
