Streptavidin-coupled Dynabeads™ are the gold standard for separation and processing of biotinylated nucleic acids, antibodies or other biotinylated ligands and target molecules. The extremely high binding affinity of the streptavidin-biotin interaction (K_d=10^-15) has been exploited in a wide range of applications. The amount of biotinylated DNA that can be immobilized depends on the fragment size. Due to steric hindrance, the binding efficiency decreases significantly when the fragment size exceeds 2 kb. A specially designed binding solution is provided as the Dynabeads™ kilobaseBINDER™ kit, which contains a patented immobilization activator, significantly increases the binding capacity of large double-stranded DNA fragments.

Applications:
The Dynabeads™ kilobaseBINDERTM kit is designed for the immobilization of long double-stranded DNA molecules (>2 kb). The product has been used for innovative applications in cell biology (e.g., construction of chromosome-mimicking chromatin beads for studying microtubule self-organization into bipolar spindles, construction of synthetic nuclei for studying nuclear RNA export in vitro, reconstruction of nuclear movement along microtubules and examination of its molecular basis in vitro. Binding Capacity:
Both the size of the molecule and the biotin labeling process affect binding capacity. Binding capacity also depends on the available space and charge interactions between the bead and the molecule and between molecules. Dynabeads™ Coated with a monolayer of recombinant streptavidin. This makes the vast majority of biotin binding sites spatially available to bind not only free biotin but also biotinylated ligand/target.

The streptavidin monolayer is covalently coupled to the beads, ensuring negligible leakage and excellent reproducibility. Using the unique binding solution provided in the kit, 70 pmole of 4 kb biotinylated DNA or 80 pmole of 10 kb fragments can be immobilized per mg of beads. Invitrogen Dynal quality control testing ensures that the following binding capacity criteria are met:
•>10-fold higher binding capacity of biotinylated 4 kb DNA fragment per mg of beads when using the binding solution compared to without the binding solution.
•>195 μg (6 pmol) biotinylated 50 kb DNA fragment/mg of beads.
In addition, the EMBL Heidelberg group that developed the binding solution reported the following results:
•Approximately 400 μg (100 pmole) biotinylated 6.2 kb DNA fragment/mg Dynabeads M-280 streptavidin.
