Unlike traditional IP procedures based on Protein A/G bead capture, this kit uses magnetic beads containing pre-immobilized anti-HA antibodies. These Pierce anti-HA magnetic beads ensure specific binding to HA-tagged protein complexes in biological samples. Because the antibodies are covalently attached to the beads, IP targets can be easily eluted and recovered without antibody contamination. The hemagglutinin (HA) peptide (YPYDVPDYA) derived from the human influenza virus HA protein is one of several fusion protein tags used for recombinant protein expression. Pierce Magnetic HA Tag IP/Co-IP Kit uses specific high affinity immobilized antibodies (clone 2-2.2.14) from bacterial and mammalian cell lysates and using the Pierce HumanIn VitroTranslation Kit HA-tagged fusion proteins were rapidly purified from prepared lysates. The beads are incubated with cell lysates containing HA-tagged proteins to capture the fusion proteins, and the beads are subsequently washed and eluted using a low pH elution buffer or a non-reducing sample buffer. The protocols and buffers are optimized to improve the efficiency of IP and co-IP reactions and enrich specific protein interaction complexes in eluted samples. Anti-HA antibodies can be used to detect HA-tagged proteins by Western blot analysis.
