Ambion™ RiboPure™ Kits are designed for rapid purification of high-quality RNA from tissue samples or cultured cells. The kit combines TRI Reagent™ with glass fiber filter column purification to obtain extremely pure RNA without residual proteins and lipids. Kit includes sufficient reagents for 50 purifications from 1–100 mg of tissue or 0.1–20 x 106 cultured cells.

RNA isolated using RiboPure™ Kits is suitable for all common downstream applications, including cDNA synthesis, real-time and end-point RT-PCR, microarray analysis, northern blotting and RNase protection assays. The RiboPure™ protocol is particularly useful for processing samples that are difficult to destroy or contain high levels of ribonucleases or other contaminants. Samples are first homogenized in TRI Reagent™, a single-phase solution containing phenol and guanidine thiocyanate, to rapidly lyse cells and inactivate nucleases. The addition of bromochloropropane (BCP) separates the homogenate into an aqueous and organic phase. RNA partitions into the aqueous phase, while DNA and proteins remain in the intermediate and organic phases. The aqueous phase was removed and passed through an RNA-bound glass fiber filter column. Wash the filter column and elute the RNA. This procedure is compatible with tissue stored in Ambion RNAlater™. Total RNA yield is typically 100–500 μg/100 mg tissue, depending on tissue type.
