Click-iT® AHA (L-azidohomoalanine) is a traditional radioactive technology for the detection of nascent proteins (35S -Methionine) offers a fast, sensitive, non-toxic and, more importantly, non-radioactive alternative. AHA is an amino acid analog that is slightly modified specifically with azide groups and can be added to cultured cells and incorporated into proteins during active protein synthesis. The assay utilizes a chemoselective ligation, or "click" reaction between an azide and an alkyne, using a Click-iT® Protein Assay Detection Kit containing TAMRA, Dapoxyl® or a biotin-alkyne to detect azide-transformed modified proteins. These reagents provide detection sensitivity in the lower femtomolar range in 1-D gel and Western blot analysis and are compatible with downstream LC-MS⁄MS and MALDI MS analysis or Multiplexed Proteomics® reagents for use with newly synthesized proteins as well as Differential analysis of total glycoproteins, total phosphoproteins or total proteins.
