CellEvent Cell Senescence Green Detection Kit consists of fluorescent probes and optimized buffers, enabling image-based detection of senescent cells . The fluorescein-labeled probe contains two galactopyranoside moieties, making it a target for β-galactosidase. Activation of β-galactosidase is commonly used as a biomarker of senescent cells. This hydrolase is located in lysosomes and converts β-galactoside to monosaccharides under acidic pH conditions. The enzymatic digestion product remains in the cell due to the covalent binding of intracellular proteins and emits a fluorescent signal, with a maximum absorption/emission wavelength of 490/514 nm.

Features of the CellEvent Cell Senescence Green Detection Kit include:
• Reliable and rapid fluorescence detection of senescent cells
• Higher sensitivity than traditional colorimetric x-gal
• Fluorescence The signal is maintained well and can be detected using the standard Alexa Fluor 488/FITC filter set
• Supports multi-channel detection—cell senescence fluorescent probes can perform multi-channel detection

Due to the limited replicative lifespan, normal cells Cell cycle arrest occurs, also known as cellular senescence. During this phase of senescence, cells remain metabolically active without cell death or division. They have a specific phenotype, including the presence of multiple nuclei, increased vacuolation, expression of pH-dependent β-galactosidase, and an enlarged and stretched appearance. Cellular senescence may play a role in tumor suppression, tumor progression, senescence, and tissue repair through multiple mechanisms.

Activation of β-galactosidase is often used as a biomarker for senescent cells. This hydrolase is located in lysosomes and converts β-galactoside to monosaccharides under acidic pH conditions. Its activity is optimal at a lysosomal pH of 4, but conventional assays are usually measured at a pH of 6. The study showed that normalized beta-galactosidase activity in senescent cells was twice that of presenescent cells, regardless of the pH used in the test.

The colorimetric substrate for β-galactosidase, 5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside (x-gal), has long been used Detection of metabolic activity of cells in vitro. However, its application is limited due to signal inconsistency, long detection time, and inability to perform multi-channel detection.

Therefore, our company has developed a sensitive β-galactosidase fluorescent substrate that can be used to detect senescent cells. The CellEvent Cell Senescence Green Probe is a fluorescein-based reagent that contains two galactopyranoside moieties, making it a specific target of beta-galactosidase. The enzymatic digestion product remains in the cell due to the covalent binding of intracellular proteins and emits a fluorescent signal, with a maximum absorption/emission wavelength of 490/514 nm.

CellEvent Cell Senescence Green Kit provides CellEvent cell senescence green probe and optimized buffer, which can be used to detect senescent cells in fixed samples. The probe can be used with other reagents for multi-pass detection and is compatible with paraformaldehyde fixation.
