Thermo Scientific Ribonuclease I (RNase I) is an endoribonuclease that passes nucleoside 2', The 3'-cyclic monophosphate intermediate preferentially hydrolyzes single-stranded RNA to nucleoside 3'-monophosphate.

The enzyme does not require metal ions to be active.

Mammalian ribonuclease inhibitors have no effect on RNase I. RNase I binds to DNA but does not degrade it. The RNase I amino acid sequence is typical of the RNase T2 family. Nonionic detergents such as Triton X-100 do not inhibit RNase I and may even slightly stimulate its activity and stabilize it against thermal inactivation. Triton X-100 or BSA (0.1 mg/mL) prevents RNase I from adhering to glass vessels when working with dilute solutions. Polyamines stimulate RNase I activity.
