Thermo Scientific phiX174 is an earlier DNA virus discovered to have a single-stranded, circular genome. The length of phiX174 DNA is 5386 nt. The DNA strand packaged into the virion is called the “+” strand. Once inside the cell, Thermo Scientific phiX174 DNA is used as a template for negative-strand synthesis, producing double-stranded DNA. Conversion of the +DNA strand to double-stranded DNA does not require any phage genes to function.
The double-stranded DNA can then be transcribed to synthesize the phage-encoded protein. Synthesis of single-stranded (+) DNA requires the phage-encoded gene A protein. DNA synthesis is initiated at the ori (+) position and proceeds in the direction indicated. Late in infection, the single strand is loop-encapsulated into new virions. The cycle is terminated by cell lysis mediated by a protein encoded by phage gene E.
