Thermo Scientific exonuclease-resistant random primers are a mixture of single-stranded random oligonucleotides that can efficiently and randomly prime different DNA synthesis reaction. These primers contain two 3-terminal phosphorothioate (PTO) modifications that are resistant to the 3'→5' exonuclease activity of proofreading DNA polymerases, such as Klenow Fragment and phi29 DNA polymerase. This primer also contains 5'- and 3'-hydroxyl termini. Exonuclease-resistant random primers are supplied as ready-to-use 20X concentrated aqueous solutions.
Features
• 3'-terminal PTO modification prevents 3'→5' exonuclease degradation and ensures efficient initiation of DNA synthesis
Applications
• Strand displacement amplification of genomic DNA (see reference 2), plasmid and phage DNA
• DNA labeling by random priming (see reference 4- 6)
