CytoVista 3D Cell Culture Clearing Reagent makes fluorescently labeled 3D cultured cells such as organoids and spheroids transparent change. This clearing process enables clear, bright 3D fluorescence imaging to depths up to 1000 µm. This reagent can also be used as an imaging solution, allowing 3D cell cultures to be stored stably for weeks with minimal changes in tissue morphology or fluorophore brightness or specificity.

< br>The CytoVista tissue and 3D cell culture clearing workflow enables the clearing of tissues and 3D cultured cells with minimal morphological changes in tissues or cells and virtually no impact on the detection sensitivity of fluorophores. With this easy-to-use protocol, whole mouse brains (8 mm sections) can be cleared in 24 hours, or 1 mm sections can be cleared in 2 hours, without the use of any special instruments or equipment.
The CytoVista clearing workflow is compatible with most fluorophores, including fluorescent proteins detected with common fluorescence imaging instruments such as widefield microscopy, high content analyzers, confocal microscopy, and light sheet microscopy. For first-time users, we recommend the CytoVista Tissue Clearing/Staining Kit (Cat. No. V11324) or the CytoVista 3D Cell Culture Clearing/Staining Kit (Cat. No. V11325). These kits contain all necessary reagents for clearing and staining with immunohistochemical techniques.
CytoVista clearing technology effectively clears tissue for 3D fluorescence imaging, but unlike other clearing technologies, it does not significantly alter tissue morphology. Very slight morphological changes such as reduction and shrinkage are observed. For precious samples, clearing is reversible and the tissue can be processed for histological studies such as H&E staining.
For tissues that are easy to clear (such as brain, lung, intestine, skin) and tissues less than 250 µm thick, processing with CytoVista Tissue Clearing Reagent is sufficient for 3D imaging. However, tissues that are thicker than 250 µm or that are difficult to clear (such as kidneys, liver, heart, and placenta) need to be processed first with CytoVista Tissue Clearing Reagent and then with CytoVista Tissue Clearing Enhancer.
CytoVista 3D Cell Culture Clearing Reagent is recommended for spheroids or organoids up to 1 mm thick.
CytoVista tissue clearing reagent is not recommended for hard tissues (such as bone, cartilage, and connective tissue).
