Using Guardian peroxidase conjugate stabilizer/diluent, commonly used 1 mg/mL antibodies and streptoavidin can be combined Peroxidase conjugates were diluted up to 100,000-fold and stored at 4°C. Most ELISA and blot applications of HRP conjugates require at least a 1000-fold dilution (to 1 μg/mL) of the commonly used 1 mg/mL stock solution. Sensitive detection systems (such as chemiluminescent substrate detection systems) often require dilutions up to 100,000-fold (10 ng/mL). Guardian solutions allow such 1X working concentration HRP conjugates to be prepared in advance and stored at 4°C for 12 months or at room temperature for 6 months. For blotting applications, it is usually best to prepare a stock concentration buffer that can be diluted 10-fold with detection buffer (containing blocking components) for end use.

A typical colorimetric ELISA experiment requires diluting 10 mL of HRP-conjugated secondary antibody 1000 to 5000 times from a 1 mg/mL stock solution. To do this, pipette 2 to 10 μL of the stock solution into a tube containing 10 mL of diluent. More sensitive detection systems—such as Western blot detection using Thermo Scientific SuperSignal West Pico, Dura, or Femto Chemiluminescent Substrate (for HRP)—require a second round of dilution to achieve the required HRP conjugate working concentration. With Guardian Stabilizer-Diluent, HRP conjugates can be pre-formulated and stored in ready-to-use concentrations or easily diluted 10- to 100-fold for final use. Larger volumes of pre-diluted stock solutions can be prepared to ensure consistency and greater accuracy in daily testing.
